I would like to understand what this number means before I repeat it anywhere.

A Medutest report on a retatrutide lot gives 98.9% purity. The supplier certificate for the same lot states 98.5%. Both documents name a reversed-phase method; neither states the same gradient.

My question is not "who is right". It is: given that those two figures were produced by different methods, what is the largest difference I should expect from method alone, and at what point does a gap stop being explainable that way?

Worth separating two things that the opening post runs together.

Excipients and their purposes: mannitol, sucrose, or other sugars in lyophilised formulations protect the peptide during drying and storage. They are not impurities; they are functional components.

Preservatives: solutions for multi-dose vials contain preservatives (benzyl alcohol, phenol, or others) to inhibit microbial growth after the vial has been entered. Single-use vials do not need preservatives.

post #4 answers the question as asked. The question underneath it is different.

Fill volume and headspace: the amount of solution in a vial and the air above it affect how much the solution is exposed to oxygen during storage. Less headspace means less oxygen exposure.

On post #2 — agreed on the reasoning, with one qualification.

Container closure: the vial material, rubber closure, and sealing all affect permeability to water and oxygen. Glass is less permeable than plastic. Rubber quality matters.

Antioxidants: compounds that prevent oxidation (which damages peptides) are sometimes included. Methionine oxidation is a common degradation pathway and antioxidants help prevent it.

Amino acid residues and formulation: the specific sequence and composition of a peptide affects what formulation conditions it tolerates. Sequences high in methionine need more antioxidant protection. Sequences prone to aggregation need different pH or additive protection.

I read post #9 twice before replying, because I had assumed the opposite.

Emulsification and suspensions: some formulations are emulsions or suspensions rather than solutions. Semaglutide formulations are solutions; some other compounds are suspensions. The physical form affects stability and administration.

Buffers and pH: the pH of a solution is buffered to maintain stability. The buffer system (phosphate, Tris, or other) is chosen to keep the solution at the pH where the peptide is most stable.

post #13 is right about the mechanism and I think understates the practical bit.

Isotonicity: an isotonic solution has the same osmotic pressure as blood. Non-isotonic solutions can cause discomfort on injection. Injectable peptide solutions are formulated to be isotonic.

Lyophilisation and reconstitution: the powder is stable for extended periods if kept cool and dry. Once reconstituted, stability decreases because the dissolved peptide is more vulnerable to oxidation and other degradation.

This follows post #18 rather than contradicting it.

I disagree with the reply above, and I think the disagreement is substantive rather than terminological.

The distinction being drawn does not survive when you look at the published data for this specific question. I would be glad to be shown wrong on this, because the version I am arguing against is more convenient.
